Combining glycoprotein blockers with fibrinolysis: a bold stroke?
نویسنده
چکیده
For the future treatment of ST-elevation myocardial infarction, fibrinolytic therapy will be the most widely used therapy, because of its simplicity, worldwide availability and relatively low cost. In comparison to primary angioplasty, which will continuously be hampered by its limited availability and inherent time delay, fibrinolytic therapy has two major drawbacks: partial efficacy and a small, but significant risk of major life-threatening bleeding. Several modifications in fibrinolytic drug design have optimized simplicity and efficacy in that 90 min patency of about 60% can be achieved with

Revisiting Beta 2 Glycoprotein I, the Major Autoantigen in the Antiphospholipid Syndrome
Beta 2 glycoprotein I (β2GPI) is a single chain 50 kDa highly glycosylated glycoprotein at an approximate concentration of 4 μM in cells. The abundance of this protein in plasma and its high state of preservation indicate the important role of this protein in mammalian. In addition, β2GPI has a particular structure in the fifth domain, and is categorized as the major antigen recognized by autoa...
متن کاملAbciximab in acute stroke
The glycoprotein (GP) IIb/IIIa receptor is a platelet-specific adhesion receptor, which is the final pathway of platelet aggregation. It is blocked by abciximab, a chimeric (murine-human) Fab fragment of a monoclonal antibody. Animal studies have shown that GP IIb/IIIa antagonists improve blood flow in the cerebral microcirculation. Angiographic studies performed in patients with acute myocardi...
متن کاملHEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY Nicked 2-glycoprotein I: a marker of cerebral infarct and a novel role in the negative feedback pathway of extrinsic fibrinolysis
2-Glycoprotein I ( 2-GPI) is proteolytically cleaved by plasmin in domain V (nicked 2-GPI), being unable to bind to phospholipids. This cleavage may occur in vivo and elevated plasma levels of nicked 2-GPI were detected in patients with massive plasmin generation and fibrinolysis turnover. In this study, we report higher prevalence of elevated ratio of nicked 2-GPI against total 2-GPI in patien...
متن کاملP 149: Effect of Glycoprotein IIb/IIIa Inhibition on Acute Ischemic Stroke Injuries
Ischemic stroke accounts for about 87 percent of all cases. It occurs as a result of an obstruction within a vessel of the brain and sudden loss of blood circulation to the corresponding area resulting in the loss of brain function. It is caused by thrombotic or embolic occlusion of an artery and is more common than hemorrhagic stroke. We know that most of the injuries after an acute ischemic s...

Recanalization methods in ischemic stroke have been progressively expanded over the past years. In addition the continuous development of specialized mechanical devices for thrombectomy a broad spectrum of new drugs has been tested: Both options, novel drugs as well as new devices, can be employed independently of each other, but in most cases a combination of the two with the standard treatmen...
